What kind of signal launches a sequence? A synth? An effects processor? (in: DSP and Plugin Devel…)

Yeah, I’m looking for a good, technical answer in plain English, if possible. I know what midi does in terms of controlling audio once it’s under way, but it would be helpful to know what kind of signal actually triggers midi events to begin, as in a daw’s sequence. I’m guessing it’s the same sort of signal that triggers a plugin, whether a software synth or an effects plugin. I wonder if it’s something other than just a midi-on thing.
